Obiectiv proiect

The project aims to develop a gel-cream disinfectant, PT1, with multiple functions: disinfection, hydration, and prevention of cutaneous dysbiosis, by combining hand hygiene functions with those of restoring the hydrolipidic barrier and using a patented biocomplex of active ingredients.

The project aims to develop a functional pilot facility for the manufacturing of skin disinfectants:

icon validation of disinfectant production technology in an industrial environment;
icon demonstrating the technology's functionality in an industrial environment through testing of physical-chemical, rheological, and dermatological properties and biocidal efficacy for the purpose of market approval.

The innovative nature of the proposed technology is determined by

icon utilizing the disinfecting and moisturizing properties of organic acids, polyphenols, and anthraquinones and capitalizing on active compounds extracted from aloe plant material, enriched with mineral bioactives;
icon formulation of the gel-cream as a stable, heterogeneous system for incorporating active ingredients through a sustainable cold emulsification process, a method suitable for thermolabile active substances;
icon the use of naturally derived lipids, such as ceramides, sterols, and free fatty acids, which have a prebiotic function and promote the maintenance of the skin microbiome, reducing transepidermal water loss while regenerating the damaged lipid barrier.

As part of the activities etapei I analysis methods were developed and successfully applied for:

A. materii prime:
  • characterisation of aloe powder (FT-IR analysis, elemental analysis, thermogravimetric analysis (TGA), inductively coupled plasma optical emission spectrometry (ICP-OES));
  • extract de aloe: total polyphenol content (TPC), expressed as gallic acid equivalent, total flavonoid content (TFC), expressed as catechin equivalent, antioxidant activity as Trolox equivalent. High-performance liquid chromatography (HPLC-DAD) analysis of phenolic acids and flavonoids, GC-MS analysis, ICP-OES analysis;
  • acid lactic;
  • chlorhexidine digluconate content.
B. active substances din produsele dezvoltate:
  • lactic acid content by HPLC-DAD;
  • chlorhexidine digluconate content, accredited method;
  • total polyphenol content;
C. antimicrobial activity of the developed product.

The Dysbioklin FD2 formula complies in terms of active substance concentrations, with a significant antimicrobial effect against Gram-positive bacteria (S. aureus) and fungi (C. albicans) and lower efficacy against Gram-negative bacteria (E. coli). The sample's antimicrobial efficacy was assessed using the diffusion method to determine antibacterial and antifungal activity. The diffusion technique was applied by inoculation on standardised discs. The method was carried out in Petri dishes on specific agar media: Mueller-Hinton medium for bacteria and Sabouraud medium for Candida albicans. This activity is supported by the bioactive components of Aloe vera, such as polyphenols and flavonoids with antioxidant and anti-inflammatory effects, anthraquinones with antimicrobial effects, moisturising and regenerating polysaccharides, amino acids, and vitamins and minerals that help maintain skin health. Through these properties, the cream supports the skin's microbial balance, hydration and regeneration.
